Abstract

Tea-tehan leaves (Acalypha siamensis) are native plants in Peninsular Malaysia, Myanmar, Thailand, Combodia, Laos and Vietnam. Acalypha siamensis is a one of the Euphorbiaceae family. They have a 0.5-5m tall shrub, branching freely with
longitudinal branches, from the leaf extract of Acalypha siamensis evaluated for antibacterial and antioxidant activity, because the leaves of Acalypha siamensis have
flavonoid compounds, tannins, saponins, and alkaloids.
This research aims to optimize the solvent in the extraction process to obtain the right solvent composition in order to get the total yield, total flavonoid levels, and the optimal level of antioxidant activity. The solvent used are methanol, acetone, and distilled water. Extracts obtained with the help of sonication to speed up the extraction
process, then calculate the amount of yield obtained. Extracts obtained were tested to calculate total flavonoids and antioxidant activity. Determination of the comparison of
the composition of the solvent using the Simplex Lattice Design method with the application Design Expert 12 Trial Version. The results showed that the composition of the solvent ratio in the teh-tehan (Acalypha siamensis) leaf extraction process gave optimal results on the yield, total
flavonoid levels, and antioxidant activity in methanol: acetone: aquades, respectively 0%: 69,280%: 30,720% with a total yield of 296,638 milligrams of quercetin equivalent
per gram of flavonoid extract, 20,002% yield, and 91,788 ppm of antioxidant activity.Keywords: Optimization, Acalypha siamensis, Simplex Lattice Design, Rendemen, Total Flavonoids, Antioxidant Activity
