Abstract
Wolfberry fruit (Lycium Ruthenicum Murray) is a plant native to central Asia and is also one of the medicinal plants which have many benefits. Wolfberry is usually used to treat many health problems, including diabetes, anemia, vision
problems, impotence disorders, and lung problems. Wolfberry can be used as an antioxidant where wolfberry contains polyphenols, polysaccharides, anthocyanins, and flavonoids which have the effect of antioxidant activity.The purpose of this research was to determine the optimal composition
comparison of the optimization of solvents in the extraction process of wolfberry fruit (Lycium ruthenicum Murray) to the number of extract yield, total flavonoid content, and antioxidant activity using the Simplex Lattice Design method. The extraction method used in this research is an ultrasonic-assisted extraction method using a composition of three different solvents, namely methanol, ethanol, and
petroleum ether. The extract obtained from the ultrasonic extraction method was then calculated the amount of yield obtained. The extract was tested to determine total flavonoid content and antioxidant activity. The etermination of the comparison of the composition of the solvent is carried out by the Simplex Lattice Design method using the Design Expert 12 Trial Version application.The results showed that the comparative composition of methanol,
ethanol, and petroleum ether solvents in the extraction process of wolfberry (Lycium ruthenicum Murray) which gave optimal results on yield, total flavonoid content, and antioxidant activity with a solvent composition ratio of 0ml
methanol, 43.50ml ethanol, and 56.50 ml petroleum ether with a predicted yield of 16.5774%, total flavonoid content of 159.021 mg quercetin equivalent per gram of extract, and antioxidant activity of 135.437 ppm.Keywords: Optimization, Lycium Ruthenicum Murray, Simplex Lattice Design, Rendemen, Total Flavonoids, Antioxidant
